Many prokaryotic mRNAs are polycistronic, meaning that a number of different proteins can be produced from a single mRNA. In contrast, eukaryotic mRNAs are monocistronic. Polycistronic mRNAs have multiple AUG initiation codons – one for each protein being translated. How does the bacterial cell distinguish between initiation codons and internal methionine codons? The answer lies in the fact that mRNAs have a Shine-Dalgarno sequence just upstream of their initiation codons. This sequence interacts with the 16S rRNA and thus helps bring the ribosome to these internal initiation codons. The internal methionine codons (the ones that are not involved in initiation) do not have upstream Shine-Dalgarno sequences. Eukaryotic protein synthesis is initiated by a different mechanism - one that involves the binding of the ribosome to the 5' cap followed by scanning down the mRNA until the first AUG is found. This identifies it as the initiation codon - all other AUG codons are then, by definition, internal. There are exceptions to the scanning mechanism for some eukaryotic mRNAs. Some mRNAs can recruit ribosomes directly to initiating AUG codons through 5' UTR elements termed internal ribosome entry sites (IRESs). Mutation of the IRES in the connexin-32 gene is a cause of Charcot-Marie-Tooth disease. Mutation of the IRES of the c-myc gene causes increased translation of this oncogene, leading to multiple myeloma (Mendell and Dietz, Cell 107:411; 2001). The figure shows the lac operon in E. coli, which is transcribed into a polycistronic mRNA that encodes the three structural proteins of the operon. Each one of the proteins is initiated at an AUG codon adjacent to a Shine-Dalgarno sequence. The lower part of the figure shows a eukaryotic mRNA, which has one and only one AUG initiation codon - the first AUG downstream of the 5' cap. * * * * Eukaryotic mRNAs have a 5' cap followed by a 5' untranslated region. Unlike prokaryotic mRNAs, they do not have a Shine-Dalgarno sequence. Eukaryotic mRNAs also have a 3' poly(A) tail.
