Model for the molecular motion associated with voltage-mediated gating * The family of inward rectifier potassium channels All members of this family share significant structural similarity but only Kir2 and Kir3 subfamilies represent channels carrying classical strongly rectifying currents. Four members of each Kir2 and Kir3 subfamilies were cloned in mammals. É Bloqueada pela Ouabaína ou estrofantina G (Strophantus gratus) * * * * * * No músculo cardíaco os íons cálcio que entram durante a fase de platô dos potencial de ação fazem o acoplamento entre a atividade mecânica e contrátil, um processo conhecido como acoplamento excitação contração. A membrana do miócito cardíaco apresentam invaginações ao longo da célula chamadas de túbulos T e ocorrem em intervalos de 2 um ao longo do eixo longitudinal da célula e coincide com a linha Z do sarcômero. * Tanto na membrana superficial e na membrana do TT são encontrados os canais de cálcio tipo L que são sensíveis a voltagem e vão se abrir durante a fase de platô. Podemos observar que o retículo sarcoplasmático está bem próximo a membrana do TT. Nessa região existe uma estrutura chamada de couplon, onde os canais de Ca tipo-L da membrana do TT e os receptores de RyR localizados na membrana do RS estão bem próximos separados apenas por uma distância de 15 nm. Esse arranjo é fundamental para garantir que o influxo de cálcio via canais tipo L ative os RyR e o cálcio estocado no RS seja liberado no citosol e promova a contração muscular. No relaxamento, é preciso fazer a receptação desse cálcio, que será bombeado de volta para o retículo por uma bomba de cálcio chamada SERCA, e vai ser transportado para o meio extracelular pelo trocador Na/Ca e pela ATPase de cálcio sarcolemal. * Essa figura mostra a compração da ULC nos músculos esquelético e cardíaco. Podemos observar que no musc esquelético a ULC é menor podendo conter até 50 RyR e como podemos notar ocorre um número maior de justaposições. Já no músculo cardíaco as ULC são maiores podendo conter de 50 a 200 ryR e com menos justaposição entre o DHPR e RyR * Fig. 1. E-C coupling in ventricular myocytes. Elas são chamadas de tríade ou diáde a depender do número de elemetos que compoe. A Triade é formada por 1 tubule T e 2 cisternas terminais do RS. E esse cálcio é liberado no citosol através de canais liberadores de cálcio, um deles é o RyR. O RyR é um canal que recebeu esse nome devido a sua capacidade de se ligar a rianodina, um alcaloide obtido de plantas. Em tecidos de mamíferos já foram identificados 3 isoformas mas a RyR2 é a mais abundante no músculo cardíaco. O RyR é um complexo macromolecular formado por várias proteínas que se organizam em volta do RyR (calsequestrina, tiadina, junctina) e podem modular a atividade do RyR. O RyR pode ser modulado pelo canal de cálcio tipo L, pelo ca, Mg, ATP....... * A FKBP tem um papel fundamental na regulação do RyR. No músculo cardíaco são encontrados 4 FKBP por RyR. Ele é responsável por promover o acoplamento do RyR com os seus respectivos vizinhos garantindo a ULC funcione de forma coordenada ou sincrônica na liberação de cálcio. Se a FKBP está dissociada do RyR, os RyR passam a funcionar como unidades isoladas perdendo o sincronismo na liberação do cálcio * Figure 5. Surface-rendered representation of triad.
