A negatively charged Asp residue on the 3rd transmembrane region (TM3), along with other charged, polar residues, allows a positively charged norepinephrine (NE) molecule to bind to the hydrophobic core of the receptor. (Click to see animation; click again for next step) 2) Binding of NE causes the third intracellular loop (i3) of the receptor to change conformation and bind to the GDP-bound αs subunit of the Gs protein. (Click to see animation; click again for next step) 3) Binding of i3 to the αs subunit of the Gs protein results in a conformation change in αs, causing GDP to dissociate and GTP to bind. (Click to see animation; click again for next step) 4) The GTP-bound αs subunit dissociates from the β subunit and from the βAR receptor and binds to adenyl cyclase (AC). (Meanwhile, norepinephrine may dissociate from the receptor, but the αs subunit can remain active for many seconds after this dissociation.) (Click to see animation; click again for next step) 5) Activated adenyl cyclase produces many molecules of cAMP from ATP. (Click to see animation; click again for next step) 6) After hydrolysis of GTP to GDP, the αs subunit returns to its original conformation, dissociates from AC (which then becomes inactive), and reforms the trimeric Gs protein complex. Binding of two cAMP molecules to each R subunit causes the C subunits to dissociate and become active. (Click to see animation; click again for next step) Nucleus Cytoplasm 2) The active catalytic subunit of PKA travels into the nucleus and physphorylates CREB dimers located on DNA promoter regions. (Click to see animation; click again for next step) 3) CREB binding protein (CBP) can now bind to the phosphorylated CREB. This activated CREB/CBP complex comes in contact with and activates RNA polymerase II downstream on the DNA strand, enabling transcription of various genes. (CREB can also activate fos, regulating transcription of slower-response genes.) (Click to see animation. Click again for next slide.) * * * GDP Asp - NE + 1) After about 5 minutes of stimulation by norepinephrine, βAR changes conformation, exposing Ser and Thr residues near the C-terminal end of the receptor. (Click to see animation; click again for next step) TM1 TM4 TM3 TM2 NE + N C β-adrenergic receptor NE + NE + TM7 Gs protein i3 loop Cytoplasm Extracellular space β-ARK 2) This change in conformation allows β-ARK to bind to the receptor and phosphorylate the Ser and Thr residues. (Click to see animation; click again for next step) β-arrestin TM5 TM6 GTP γ αs β 3) Phosphorylation of these residues allows β-arrestin to bind. The βAR i3 loop changes conformation and uncouples from the Gs protein. (Click to see animation; click again for next step) cAMP cAMP cAMP 4) Inactivation of the Gs protein results in the lowering of cAMP levels and the subsequent reduction of gene transcription through the CREB pathway. (Click to see animation; click again for next step) AP2 5) The β-arrestin/receptor complex binds to membrane-bound AP2, which is responsible for endocytosis of the receptor into clathrin-coated pits. After about 24 hours, some of the receptors are returned to the cell membrane. (Click to see animation.)
