As NETs são compostas basicamente por cromatina nuclear descondensada juntamente com proteínas, onde a maioria delas vem dos grânulos dos neutrófilos e as outras proteínas são citoplasmáticas e nucleares (histonas). Para confirmar que os neutrófilos respondem ao BRSV, examinamos neutrófilos bovinos isolados expostos a PMA, BRSV ou ambos in vitro A) Desenho de pulmão bovino indicando áreas onde amostras de tecido foram coletadas e criopreservadas usando nitrogênio líquido. Coletamos pelo menos três amostras de cada lobo. Resultados 03. Todos os bezerros desenvolveram doença clínica e excreção viral Todos os bezerros tiveram pontuações clínicas mais altas a partir do dia 3 Grupo 3 (placebo) - pontuações clínicas altas - doença mais grave Grupo 6 (terapia combinada a partir do dia 3) teve a pontuação clínica mais baixa Ibuprofeno aumentou a excreção viral e FPI diminuiu a excreção viral Resultados Escores clínicos e patológicos Escores histopatológicos mais altos - placebo em todo o tratamento e ibuprofeno a partir do dia 5 Escores mais reduzidos - terapia de combinação com ibuprofeno e FPI iniciada no dia 3 Escores clínicos e patológicos Resultados observamos redes semelhantes a teias de DNA extracelular em torno de neutrófilos bovinos desnaturantes desafiados com BRSV e PMA Redes semelhantes a teias de DNA extracelular em torno de neutrófilos Análise de NETs in vitro Resultados NETs mais abundantes em neutrófilos estimulados por PMA Revestidas com elastase e citH3 - formação de NETs Imagens imunofluorescentes representativas de amostras de citologia de bezerros saudáveis. As células foram fixadas, permeabilizadas e coradas para histona citrulinada H3 (citH3, vermelho), elastase de neutrófilos (NE, verde) e DNA (azul). NETs foram identificados por co-localização de citH3 e NE. As células foram identificadas como neutrófilos com base em núcleos lobulados Pulmões - apresentavam lesões macroscópicas com distribuição cranioventral médio e caudal Patologia e NETs induzidas por BRSV em neutrófilos bovinos in vivo Resultados Pneumonia bronco-intersticial, bronquiolite necrosante, formação de sincícios, hiperplasia de pneumócitos tipo II e alveolite exsudativa ou proliferativa NETose generalizada e deposição de NETs - coloração positiva de CitH3 e NE Deposição de NETs em áreas intraluminais e intersticiais As amostras do grupo placebo continham significativamente mais coloração de citH3 e NE C) e D): distribuição de NETs nos lobos pulmonares cranial, médio e caudal com alta Netose visualizada no lobo cranial para histonas citrulinadas e elastase de neutrófilos. Dados representados como uma porcentagem do total de três repetições. n = 6 por grupo, Vermelho – lobo craniano, Amarelo – lobo médio, Azul – lobo caudal. B) Uma imagem imunofluorescente representativa de tecido pulmonar bovino congelado. Os tecidos foram fixados, permeabilizados e corados para histona citrulinada H3 (citH3-verde) e elastase de neutrófilos (NE-red). O DNA foi corado com DAPI-blue. Observe a expressão intracelular de citH3 (cabeças de seta) e NE (setas) em neutrófilos, e liberação de NETs decorados com NE e citH3 (circulado). O BRSV pode induzir a formação de NETs tanto em vitro quanto in vivo. A formação de NETs foi observada nos pulmões, sugerindo que os NETs contribuem para a imunopatologia na doença do VSR. Ibuprofeno e FPI, em média diminuem a gravidade clínica da doença e NETosis, mas deve ser usado em conjunto e não como monoterapia. A combinação de ambas as drogas foi eficaz no 3 e 5 após a infecção com início precoce levando a melhores resultados. Este trabalho ressaltaa necessidade de mais estudos sobre compostos que influenciam NETs. Também especula-se que há necessidade de diagnóstico precoce de doenças exacerbadas por NETs para que a terapia seja eficaz, pois é u trabalho contra o tempo em competição com uma resposta inflamatória.
