O hipotálamo e a glândula pituitária têm uma relação muito próxima, formando o eixo hipotálamo-hipofisário. O hipotálamo secreta hormônios que estimulam ou inibem a secreção de hormônios pela glândula pituitária. A glândula pituitária, por sua vez, secreta hormônios que regulam o funcionamento de outras glândulas endócrinas no corpo. A hipófise anterior secreta hormônios como o hormônio do crescimento, hormônio estimulante da tireoide, hormônio adrenocorticotrófico, hormônio folículo-estimulante e hormônio luteinizante. A hipófise posterior secreta hormônios como a ocitocina e a vasopressina.
