A secreção e liberação do leite são controladas hormonalmente. Durante a amamentação, a estimulação dos receptores sensíveis presentes na papila mamária pela ocitocina é necessária para a liberação do leite. A informação sobre o processo de sucção é transmitida ao hipotálamo por meio de fibras nervosas sensíveis. Essa informação inibe a produção de dopamina e estimula a produção de prolactina. A liberação do leite ocorre com a ajuda dos movimentos de contração das células mioepiteliais em resposta à liberação de ocitocina. O hormônio prolactina é responsável pela produção da maior parte do leite consumido pelo bebê durante a amamentação.
