The temperature affects the speed of chemical reactions, being higher the speed the higher the temperature, until a certain point called optimal temperature, after which the reaction slows down due to enzyme denaturation.
Enzymes have an optimal pH in which their speed is maximum. The amount of H+ ions affects enzymes in various ways, such as altering the ability to ionize their active sites and changing the bonds that sustain the quaternary and tertiary structure of enzymes.
The initial velocity (V0) is directly proportional to the amount of enzyme, considering that there is an excess of substrate. When the amount of substrate saturates the active sites of the enzymes, only enzyme-substrate complexes (ES) exist, and the reaction velocity stabilizes, reaching the maximum reaction velocity (Vmax).
