Prof. Calçada 2019 Estereoisomeria Introdução Moléculas construídas como se uma fossem uma a imagem da outra num espelho plano, e que não se sobrepõem, são diferentes uma da outra, ou seja são isômeras. Esse tipo de isomeria é chamada de enantiomeria (“isomeria óptica”). Morrison and Boyd: “We shall make no use of the term optical isomer, since it is hard to define – indeed, is often used undefined – and of doubtful usefulness”. 1. Condição para ocorrer A isomeria óptica é ATRIBUTO DA ASSIMETRIA MOLECULAR 2. Características dos “isômeros ópticos” (melhor: para o PAR DE ENANTIÔMEROS): ü Têm propriedades químicas e físicas idênticas*. ü Têm propriedades fisiológicas diferentes. ü São chamados de isômeros opticamente ativos (IOA), pois, interagem com a luz polarizada de forma diferente. Rotacionam o plano da luz polarizada: Para a direita: são chamados de dextrogiro (d) ou (+). Para a esquerda: são chamados de levogiro (!) ou (-). * exceto a interação com a luz polarizada (“meio quiral”). 3. C* é o carbono assimétrico ou quiral ou centro de assimetria. M ATENÇÃO: ter C* não significa ter isômeros ópticos!!! ATENÇÃO: se a molécula tiver UM carbono assimétrico ela será quiral e terá dois isômeros ópticos: d e !�(PAR DE ENANTIÔMEROS) 4. Mistura racêmica É o nome dado a uma mistura equimolar dos isômeros d e !� Essa mistura não desvia a luz polarizada e é opticamente inativa (também chamada de “isômero opticamente inativo” – IOI). 5. Quantidade de isômeros Quantidade de isômeros Para n C* DIFERENTES temos: IOA = 2n e IOI = 2n/2 IOA é total de isômeros d e ! que podemos obter. Como ocorrem sempre em pares de enantiômeros, IOI é metade de IOA. M ATENÇÃO Se a molécula tiver 2 C* diferentes ela terá 4 IOA. Porém, são dois pares de enantiômeros. Um enantiômero é isômero óptico apenas de sua imagem no espelho. Quando duas molécula são isômero espacial uma da outra, MAS NÃO formam um par de enantiômeros elas NÃO SÃO isômeros ópticos – são chamadas de DIASTEREOISÔMEROS (muitos autores usam diasteroisômeros). Um bom exemplo de diastereoisômeros são: D – glicose e D – galactose (são isômeros opticamente ativos, mas uma não é enantiômera da outra – em outras palavras, uma não é “isômera óptica” da outra). Amusingly, the fi rst assignment of a three-dimensional struc- ture to a chiral molecule was a guess made more than a century ago by the sugar chemist Emil Fischer.* He did so in a paper in 1891 to simplify the depiction of the compli- cated stereochemical relationships of grape sugar (glucose) to other sugars, a topic that we will visit in Chapter 24. His choice (by chemical correlation) assigned the naturally occurring dextrorotatory enantiomer of 2,3-dihydroxypropanal (glyceraldehyde) the three-dimensional structure labeled d-glyceraldehyde. The label “d” does not refer to the sign of rotation of plane-polarized light but to the spatial arrange- ment of the substituent groups. The Fischer Stencil is shown as C ≥ !# ´ A Fischer (Real Life 5-2) projection is a simplifi ed way of depicting tetrahedral carbon atoms and their substituents in two dimensions. With this method, the molecule is drawn in the form of a cross, the central carbon being at the point of intersection. The horizontal lines signify bonds directed toward the viewer; the vertical lines are pointing away. Hashed- wedged line structures have to be arranged in this way to facilitate their conversion into Fischer projections. As luck (for the discoverers) would have it (see also Problem 64), an enzyme in the body, a-methylacyl-CoA racemase, converts the inactive (R) to the (S) isomer to the tune of 63%, thus adding value to the racemic mixture.
