Antidepressants block the reuptake of neurotransmitters, leading to an increase in their concentration in the synaptic cleft.
The increase in neurotransmitter concentration causes desensitization/downregulation of presynaptic autoreceptors, leading to more neurotransmitter release.
The increase in neurotransmitter concentration also causes desensitization/downregulation of postsynaptic receptors, reducing side effects.

O mecanismo de ação dos antidepressivos é bloquear a recaptação dos neurotransmissores, levando a um aumento na sua concentração na fenda sináptica. Esse aumento na concentração de neurotransmissores causa dessensibilização/downregulation dos autoreceptores pré-sinápticos, levando a mais liberação de neurotransmissores. O aumento na concentração de neurotransmissores também causa dessensibilização/downregulation dos receptores pós-sinápticos, reduzindo os efeitos colaterais. Além disso, os antidepressivos causam downregulation dos receptores ß1 e alfa 2-adrenérgicos após o tratamento crônico e, em alguns casos, também pela eletroconvulsoterapia - noradrenalina e dopamina.
